黃花石斛(Dendrobium catenatum Lindl.)為臺灣原生之春石斛,其花朵具觀賞價值、假球莖含有多種機能性成分,長期被作為中藥材使用,有潛力發展為觀光景點與特用作物,然而缺乏穩定的種苗繁殖技術。本研究擬利用組織培養建立一套完整之分生苗生產流程。將黃花石斛新芽及高芽切成帶節莖段培養於含9 mg.L-1 BA的培養基中,高芽培植體較新芽汙染率低、培養10週後可增殖出芽體或PLB,芽長芽增殖出之小苗繼代後生長良好、易發根,小苗從節間分切微扦插後可繼續增殖芽體,其基部切段具有最高之增殖率。將已發根組培苗馴化1個月後出瓶種植,高度7公分以上的中、大苗無論拆成單芽或維持多芽都可有高存活率,惟出瓶1個月內栽培環境需保持高濕度避免葉尖焦枯,植株高度小於7公分的小苗拆成單芽則容易黃化、降低存活率。本研究建立之黃花石斛微體繁殖技術有應用於種苗商業量產之潛力。
Dendrobium catenatum Lindl. is a native species of noble type Dendrobium in Taiwan. The flowers had ornamental values and the pseudobulbs contain many functional compounds. It has long been used as a traditional Chinese medicinal herb and has the potential to be developed into a tourist attraction and a specialty crop. However, stable seedling propagation techniques are still lacking. This study aimed to establish a complete production process of vegetative seedling through tissue culture. New shoots and keiki were cut into stem segments with nodes and were then incubated in a medium containing 9 mg·L-1 BA. Explants from keiki had lower contamination and could proliferate shoots or protocorm-like bodies (PLBs) after 10 weeks. The plantlets derived from shoots proliferation grew well and rooted easily. The plantlets could further propagate new shoots by micro-cutting through recutting between the nodes, with basal segments showing the highest proliferation rate. After one month of acclimatization, the in vitro seedlings could be transplanted ex-vitro. Medium and large seedlings with height > 7 cm exhibited high survival rates whether divided into single shoots or remained multiple shoots. However, within the first month after ex-vitro, high environment humidity should be maintained to prevent leaf tip necrosis. In the contrast, the small seedlings with height < 7cm exhibited yellowing and lower survival rates when divided into single shoots. The micropropagation technique for Dendrobium catenatum established in this study had the potential to apply in seedling mass production in commercial.
